Received for publication 17 January 1979 and in revised form 23 July 1979. reducing incorporation into the deoxyribonucleotide pool. In contrast, deoxyadenosine plus EHNA did not show this differential inhibition of [3H]uridine incorporation into DNA, and the alteration in [3H]cytidine incorporation into nucleotide pools was less impressive. These data show an association between accumulation of dATP or dGTP and a primary inhibition of DNA synthesis, and they provide support for ribonucleotide reductase inhibition as the mechanism responsible for deoxyguanosine toxicity. Deoxyadenosine toxicity, however, appears to result from another, or perhaps a combination of, molecular event(s).

On the mechanism of inhibition of deoxyribonucleic acid synthesis in Ehrlich ascites tumor cells by deoxyadenosine in vitro.

It has previously been shown that the incorporation qf both C14-formate into DNA1 thymine and P32-orthophosphate into DNA phosphate of the hypotetraploid strain of Ehrlich ascites tumor cells in vitro is strongly inhibited by deoxyadenosine.2 It was, furthermore, shown that this inhibition is not due to a prevention of the de novo synthesis of acid-soluble thymine compounds. The incorporation o...

The effect of the 2-amino group of 7,8-dihydro-8-oxo-2′-deoxyguanosine on translesion synthesis and duplex stability

Replication of DNA containing 7,8-dihydro-8-oxo-2'-deoxyguanosine (OxodG) gives rise to G --> T transversions. The syn-isomer of the lesion directs misincorporation of 2'-deoxyadenosine (dA) opposite it. We investigated the role of the 2-amino substituent on duplex thermal stability and in replication using 7,8-dihydro-8-oxo-2'-deoxyinosine (OxodI). Oligonucleotides containing OxodI at defined ...

Evidence that binding of 7,12-dimethylbenz(a)anthracene to DNA in mouse embryo cell cultures results in extensive substitution of both adenine and guanine residues.

Primary mouse embryo cell cultures were grown in the presence of [14C]guanine, labeling primarily deoxyguanosine residues in the cellular DNA, or in the presence of [14C]adenine, labeling both deoxyguanosine and deoxyadenosine residues in the cellular DNA. These cultures were subsequently exposed to 7,12-[3H]dimethylbenz(a)anthracene for 24 hr. The DNA was isolated and hydrolyzed to deoxyribonu...

Nucleolar reconstruction was retarded in cells after treatment with adenosine, deoxyadenosine, and certain combinations of agents including deoxyribonueleosides. A nucleolar change resembling that induced by adenosine or 6-thioguanine was produced in interphasic cells by a combina tion of deoxyadenosine, deoxyguanosine, and deoxycy...

Interaction of low-energy electrons with the purine bases, nucleosides, and nucleotides of DNA.

The authors report results from computational studies of the interaction of low-energy electrons with the purine bases of DNA, adenine and guanine, as well as with the associated nucleosides, deoxyadenosine and deoxyguanosine, and the nucleotide deoxyadenosine monophosphate. Their calculations focus on the characterization of the pi* shape resonances associated with the bases and also provide g...
